Dias,Decivaldo S.; Coelho,Milton V..

Histomorphometric analysis of the response of rat skeletal muscle to swimming, immobilization and rehabilitation BJMBR
Nascimento,C.C.F.; Padula,N.; Milani,J.G.P.O.; Shimano,A.C.; Martinez,E.Z.; Mattiello-Sverzut,A.C..
The objective of the present study was to determine to what extent, if any, swimming training applied before immobilization in a cast interferes with the rehabilitation process in rat muscles. Female Wistar rats, mean weight 260.52 ± 16.26 g, were divided into 4 groups of 6 rats each: control, 6 weeks under baseline conditions; trained, swimming training for 6 weeks; trained-immobilized, swimming training for 6 weeks and then immobilized for 1 week; trained-immobilized-rehabilitated, swimming training for 6 weeks, immobilized for 1 week and then remobilized with swimming for 2 weeks. The animals were then sacrificed and the soleus and tibialis anterior muscles were dissected, frozen in liquid nitrogen and processed histochemically (H&E and mATPase)....

Interaction between human cytomegalovirus UL136 protein and ATP1B1 protein BJMBR
Cui,Xin; Sun,Zheng-Rong; Ren,Gao-Wei; Wang,Gui-Li; Qi,Ying; Ma,Yan-Ping; Ruan,Qiang.
Interplay between the host and human cytomegalovirus (HCMV) has a pivotal role in the outcome of infection. A region (referred to as UL/b’) present in the Toledo strain of HCMV and low passage clinical isolates contains 19 additional genes, which are absent in the highly passaged laboratory strain AD169. Products of the UL/b’ genes may determine the manifestations of HCMV infection in vivo. However, little is known about the host factors, which interact with UL/b’ proteins. This study was conducted to investigate the function of the HCMV UL136 protein. By yeast two-hybrid screening, the β1 subunit of the host Na+/K+-ATPase (ATP1B1) was identified to be a candidate protein, which interacts with the HCMV UL136 protein. The interaction was further evaluated...

Mitochondrial ATPase activity and membrane fluidity changes in rat liver in response to intoxication with Buckthorn (Karwinskia humboldtiana) Biol. Res.
Cid-Hernández,Margarita; Ramírez-Anguiano,Ana C; Ortiz,Genaro G; Morales-Sanchez,Eddic W; González-Ortiz,Luis J; Velasco-Ramírez,Sandra F; Pacheco-Moisés,Fermín P.
BACKGROUND: Karwinskia humboldtiana (Kh) is a poisonous plant of the rhamnacea family. To elucidate some of the subcellular effects of Kh toxicity, membrane fluidity and ATPase activities as hydrolytic and as proton-pumping activity were assessed in rat liver submitochondrial particles. Rats were randomly assigned into control non-treated group and groups that received 1,1.5 and 2 g/Kg body weight of dry powder of Kh fruit, respectively. Rats were euthanized at day 1 and 7 after treatment RESULTS: Rats under Kh treatment at all dose levels tested, does not developed any neurologic symptoms. However, we detected alterations in membrane fluidity and ATPase activity. Lower dose of Kh on day 1 after treatment induced higher mitochondrial membrane fluidity than...

Phylogenetic relationships of the European newts (genus Triturus) tested with mitochondrial DNA sequence data Naturalis
Zajc, I.; Arntzen, J.W..
European newts (genus Triturus) are widely studied, but their phylogeny is not yet unambiguously resolved. Fragments of mitochondrial DNA experiencing different rates of evolution (the ATPase and 12S rRNA genes) were sequenced in order to test a phylogenetic hypothesis derived from biochemical and behavioral data. Well-supported branches of the existing phylogeny gained support in our study. The monophyletic origin of the hypothesized T. boscai — T. italicus clade remained ambiguous, whereas strong support was gained for the sister-taxon relationship of T. vulgaris and T. montandoni. The position of T. vittatus as a sister taxon to the T. marmoratus species group was also supported. The phylogenetic position of T. alpestris could not be clarified. With an...

Phylogeography and conservation genetics of the Amazonian freshwater stingray Paratrygon aiereba Müller & Henle, 1841 (Chondrichthyes: Potamotrygonidae) Neotropical Ichthyology
Frederico,Renata G.; Farias,Izeni P.; Araújo,Maria Lúcia Góes de; Charvet-Almeida,Patricia; Alves-Gomes,José A..
The family Potamotrygonidae is monophyletic comprising three genera: Paratrygon Duméril, Potamotrygon Garman and Plesiotrygon Rosa, Castello & Thorson. The distribution of most species in this family is restricted to a single basin or fluvial system. Only Potamotrygon motoro, Potamotrygon orbignyi and Paratrygon aiereba are found in more than one river basin. In this study we investigate genetic structuring of Paratrygon aiereba, from five rivers of the Amazon region: Negro, Solimões-Amazon-Estuary system, Tapajós, Xingu and Araguaia. Sixty-three individuals were sequenced for ATPase 6, and a representative subsample of 27 individuals was sequenced for COI. The COI dataset analysis indicated that Paratrygon is sister to all other potamotrygonid...

Phylogeography of Hypostomus strigaticeps(Siluriformes: Loricariidae) inferred by mitochondrial DNA reveals its distribution in the upper Paraná River basin Neotropical Ichthyology
Borba,Rafael Splendore de; Zawadzki,Claudio Henrique; Oliveira,Claudio; Perdices,Anabel; Parise-Maltempi,Patrícia Pasquali; Alves,Anderson Luis.
In this study, phylogenetic and phylogeographic analyses of populations identified as Hypostomus strigaticeps from the upper Paraná River basin were conducted in order to test whether these different populations comprises cryptic species or structured populations and to assess their genetic variability. The sequences of the mitochondrial DNA ATP sintetase (subunits 6/8) of 27 specimens from 10 populations (one from Mogi-Guaçu River, five from Paranapanema River, three from Tietê River and one from Peixe River) were analyzed. The phylogeographic analysis showed the existence of eight haplotypes (A-H), and despite the ancestral haplotype includes only individuals from the Tietê River basin, the distribution of H. strigaticeps was not restricted to this...

Simultaneous measurement of mitochondrial respiration and ATP production in tissue homogenates and calculation of effective P/O ratios ArchiMer
Salin, Karine; Villasevil, Eugenia M.; Auer, Sonya K.; Anderson, Graeme J.; Selman, Colin; Metcalfe, Neil B.; Chinopoulos, Christos.
The use of tissue homogenate has greatly aided the study of the functioning of mitochondria. However, the amount of ATP produced per oxygen molecule consumed, that is, the effective P/O ratio, has never been measured directly in tissue homogenate. Here we combine and refine existing methods previously used in permeabilized cells and isolated mitochondria to simultaneously measure mitochondrial ATP production (JATP) and oxygen consumption (JO(2)) in tissue homogenate. A major improvement over existing methods is in the control of ATPases that otherwise interfere with the ATP assay: our modified technique facilitates simultaneous measurement of the rates of uncorrected ATP synthesis and of ATP hydrolysis, thus minimizing the amount of tissue and time needed....
